We have a GUI for one of the applications I work on that's written almost entirely in Python + PyQt. Everything voxmea said is true, for all good and all ill.

Use the binary installer for Windows from the Python website. The next step is installing the Python bindings, which can be obtained from the download page at the PyQt website.
